This article discusses the effects of stress and age on the paraventricular nucleus (PVN) of the hypothalamus. This study looks prepubertal animals ( 25 – 28 days of age) to see if there was a delayed response in ACTH and corticosterone compared to adults ( 165 days of age) , after chronic stress. The researchers also looked at differential activation of corticotrophin- releasing hormone (CRH) neurons in the PVN of the hypothalamus. The researchers wanted to look into more possible differences in PVN structures and functions in prepubertal and adult male rats. The study consisted of three different experiments in which the experimenters were blind to the experimental conditions of each subject. The first experiment was conducted to obtain data on the prepubertal and adult PVN. The second experiment was performed to determine the number of parvocellular neurosecretory neurons in the prepubertal and adult PVN. The third and final experiment was conducted to establish whether acute or repeated stress alters CRH mRNA expression differently in the PVN before and after pubertal development. The results found that between adult and prepubertal development , PVN volume and somal size and cell numbers are similar in the parvocellular and magnocellular subdivision of the PVN. The researchers also found that neither age nor stress significantly influences CRH expression. The results show that age and experience with stress work together to modulate CRH expression in the PVN.
